The MGH Institute campus comprises parts of four buildings in and around the historic Charlestown Navy Yard in Boston.

The Catherine Filene Shouse Building at 36 First Avenue is the graduate school's main building which houses most of the classrooms, labs and departmental offices. Departmental offices for the School of Nursing, the Department of Communication Sciences and Disorders, Department of Occupational Therapy and Physician Assistant Studies are all in the Shouse Building. The Office of Information Technology and Office of Facilities & Operations are also located there, on the fourth floor.

The Executive Administrative offices, including the Offices of the President, Finance & Administration, Office of the Provost, Development, Human Resources, Communication & Marketing are across the street, on the second and third floors of Building 34.

Down one block in Building 39, on the ground floor, are the Office of Student Affairs and departmental offices for the School of Nursing.

In January 2012 the Institute expanded by 21,000 square feet taking over the top floor of 2 Constitution Center, just outside the Navy Yard and directly overlooking the USS Constitution, more commonly known as “Old Ironsides.” The addition includes new and expanded physical therapy labs, a 102-seat active learning classroom with state-of-the-art technology, the new Functional Living Lab for occupational therapy, and lounge and study areas that students from nursing, occupational and physical therapy, and speech-language pathology share.

In 2013, the school expanded into two new buildings, giving it a total of 130,000 square feet in six buildings. The expansion includes office on the ground floor of One Constitution Center for the Department of Physical Therapy, and 14,000 square feet in the new Center for Health & Rehabilitation Research in Building 79/96 to accommodate faculty researchers who are leading the Institute's growing focus on research, with research funding surpassing $2 million for the first time in its history.
